越来越多的证据表明,环状RNA(circRNA)作为新型的非编码RNA,在癌变和肿瘤进展中起着至关重要的作用。然而,人们很大程度上尚不清楚circRNA在透明细胞肾细胞癌(ccRCC)中的功能和分子机制。该研究在临床样品、ccRCC细胞和体内动物模型中鉴定了circAGAP1的表达和功能。通过荧光原位杂交,RNA免疫沉淀和荧光素酶分析研究了circAGAP1的分子机制。结果显示,与相邻的非肿瘤组织相比,ccRCC组织中的circAGAP1(circ0058792)表达明显上调。此外,circAGAP1的表达与ccRCC患者的肿瘤大小、核分级和临床分期密切相关。机理研究表明,胞质circAGAP1在RNA诱导的沉默复合物中靶向miR-15-5p。另外,在ccRCC中miR-15-5p表达下调。萤光素酶报告基因检测结果表明,E2F转录因子3(E2F3)是miR-15-5p的靶标,而ccRCC中上调的E2F3表达与circAGAP1正相关。此外,在体外和体内,miR-15-5p模拟物可减轻circAGAP1的促肿瘤功能。我们的结果表明,circAGAP1通过刺激miR-15-5p发挥其作为致癌内源性RNA(ceRNA)的致癌功能,从而促进E2F3的表达。靶向circAGAP1可能是ccRCC中一种新的有吸引力的治疗策略。
