1、环状RNA编码的致癌性E-钙粘着蛋白变体通过激活EGFR-STAT3信号传导促进胶质母细胞瘤致瘤性

激活的EGFR信号驱动50%的胶质母细胞瘤(GBM)致瘤性。但是,已证明EGFR靶向疗法在治疗GBM患者方面无效,这表明存在多余的EGFR激活。环状RNA是共价闭合的RNA转录本,涉及各种生理和病理过程。在本文中,我们报告了GBM的EGFR信号转导的额外激活机制,该机制由新发现的分泌E-钙粘蛋白蛋白变体(CE-Cad)主导,该变体由环状E-钙粘蛋白(circ-E-Cad)RNA通过多轮滚环翻译编码。 C-E-Cad在GBM中过表达,并促进神经胶质瘤干细胞的致瘤性。 C-E-Cad通过使用独特的14个氨基酸的羧基末端与EGFR CR2结构域结合,激活了独立于EGF的EGFR,从而维持了神经胶质瘤干细胞的致瘤性。值得注意的是,抑制C-E-Cad可以显著增强GBM中治疗性抗EGFR策略的抗肿瘤活性。我们的研究结果揭示了C-E-Cad在刺激EGFR信号传导中的关键作用,并为治疗EGFR驱动的GBM提供了有希望的方法。

骨肉瘤是青少年中最常见的原发性恶性骨肿瘤。尽管化疗法与手术相结合可以改善某些患者的预后,但是化疗耐药性仍然是骨肉瘤治疗中的巨大障碍。越来越多的证据表明,环状RNA(circRNA)参与了癌症的发展和转移,但它们在骨肉瘤中的特定作用仍未得到充分描述。在这项研究中,我们进行了circRNA深度测序,并从人骨肉瘤细胞系组(143B,HOS,SJSA和U2OS)和人成骨细胞hFOB 1.19(对照)中鉴定了88种不同的circRNA。我们发现circCAMSAP1(也称为hsa_circ_0004338)在人骨肉瘤组织和细胞系中显著上调,并且与骨肉瘤的发展呈正相关。沉默circCAMSAP1可有效抑制骨肉瘤细胞的生长、凋亡、迁移和侵袭。此外,我们验证了circCAMSAP1通过circCAMSAP1/miR-145-5p/FLI1途径在骨肉瘤的发生中起作用。 FLI1促进骨肉瘤的肿瘤发生,而miR-145-5p抑制FLI翻译。 circCAMSAP1直接在细胞质中隔离miR-145-5p,并抑制其抑制骨肉瘤肿瘤发生的活性。此外,在大鼠中检查并验证了circCAMSAP1上调的调节作用。总之,我们的发现提供了证据,证明circCAMSAP1充当“ microRNA海绵”,并提出了人类骨肉瘤的新治疗靶标。
