已经证明某些circRNA可以编码肽,即这些环状RNA应重新定义为编码转录本。我们提出蛋白质诱饵假说来描述circRNA编码的蛋白质的功能机制。circRNA编码的蛋白具有独特的特征,例如与mRNA编码的同工型具有高序列同源性,且长度短,从而使其可以作为诱饵与同类蛋白的伴侣竞争性结合。蛋白质诱饵假说可用于解释各种生物学现象,并针对不同的病理生理事件设计治疗性干预措施。最近的研究表明,环状RNA(circRNA)编码的大量蛋白质可能在癌症发展中发挥作用。但是,我们对这组蛋白质及其所涉及的功能机制的理解仍然存在很大差距。因此,我们提出了蛋白质诱饵假说,该假说指定了circRNA编码的蛋白质与它们的同源线性剪接的蛋白质同工型竞争结合分子,从而阻止了适当的同工型功能。该假设可能会扩展我们对circRNA编码蛋白功能机制的理解,并被证明有助于阐明人类发育,生理学和疾病的潜在机制,同时也有助于药物开发。

大肠癌(CRC)是全世界人民的恶性疾病之一,主要是由于缺乏有效的医疗干预措施以及对CRC发病机理的机制理解不足所致。非编码RNA(ncRNA)是一类异源转录本,不具有编码蛋白质的能力,但具有调节蛋白质编码基因表达的能力。自噬是一种进化上保守的分解代谢过程,其中细胞质内容物被递送至细胞溶酶体进行降解,从而导致细胞成分的周转并产生细胞功能所需的能量。越来越多的证据表明,ncRNA,自噬以及ncRNA与自噬的串扰在CRC的起始、进展、转移、复发和治疗抗性中起着复杂的作用,这赋予ncRNA和自噬作为CRC临床生物标志物和治疗靶点。在这篇综述中,我们试图描述ncRNA在CRC发病机理中的复杂作用,主要包括miRNA,lncRNA和circRNA,特别是着眼于ncRNA在CRC相关自噬中的调控作用,试图阐明复杂的病理机制,涉及ncRNA和自噬,负责CRC肿瘤的发生和发展,从而为临床环境中基于ncRNA和自噬的CRC治疗策略奠定基础。

胰腺癌是预后较差的恶性肿瘤之一。 胰腺癌发生和恶性进展的分子机制尚未完全阐明。 几个关键的信号传导途径,如Notch,Wnt和Hedgehog途径,对于促进胰腺癌的发生很重要。 近来,非编码RNA,特别是环状RNA(circRNA),已被表征为参与胰腺癌的发展。 因此,在这篇综述文章中,我们描述了circRNA与胰腺癌预后之间的关联。 此外,我们讨论了circRNA如何参与胰腺癌细胞过程的调控,包括增殖、凋亡、细胞周期、迁移、侵袭、EMT、转移、血管生成、耐药性和免疫逃逸。 此外,我们提到了几种化合物可以调节circRNA的表达,这表明通过化合物靶向circRNA可能有助于治疗胰腺癌患者。
