肝硬化是公认的发展为肝细胞癌(HCC)的危险因素。很少有研究报告与癌旁异常增生性结节(DN)样本相比HCC样本中circRNA的表达情况。该研究使用Arraystar Human circRNA Array结合激光捕获显微切割技术(LCM)分析与癌旁DN样本相比在HCC样本中circRNA的表达情况。然后,使用体外和体内HCC模型确定关键circRNA在HCC进展和治疗敏感性中的作用和机制。研究结果发现circMEMO1在HCC样本中显著下调,并且circMEMO1的水平与HCC患者的OS和无病生存率(DFS)密切相关。机理分析表明,circMEMO1可以充当miR-106b-5p的海绵来调节TCF21的启动子甲基化和基因表达,从而调节HCC进程,而miR-106b-5p靶向TET基因家族并增加5hmC水平。更重要的是,circMEMO1可以增加HCC细胞对索拉非尼治疗的敏感性。该研究确定circMEMO1可以促进TCF21的去甲基化和表达,并且可以被认为是肝癌进展中至关重要的表观遗传修饰因子。

2、健康和疾病的人类胎盘中RNA景观

胎盘是母亲与胎儿之间的链接,功能不足会导致短期和长期的不良健康。大多数大规模RNA-Seq数据集均不含胎盘。因此,我们分析了302个人胎盘的lnc-RNA和小RNA(每个样品分别约101和2000万个读数),其中包括94例子痫前期(PE)和56例胎儿生长受限(FGR)。胎盘转录组在50个人体组织中的复杂度排名第七低:271个基因占所有读数的50%。我们鉴定了多个环状RNA,并通过circRNA拼接位点处的Sanger测序验证了其中的6个。使用大规模质谱数据集,我们发现了两个环状RNA通过翻译产生多肽的有力证据。我们还确定了新的piRNAs,它们聚集在Chr1和Chr14上。 PE和FGR与mRNA,lincRNA和circRNA的多重和重叠差异相关,但小RNA的一致性差异较小。在PE和FGR中差异表达的三个蛋白质编码基因中,一个编码分泌的蛋白质FSTL3(类卵泡抑素样3)。孕妇血清FSTL3的升高预示着随后的PE和FGR。为了帮助可视化胎盘转录组数据,我们开发了一个Web应用程序(https://www.obgyn.cam.ac.uk/placentome/)。
