1、 M2巨噬细胞衍生的小细胞外囊泡的CircUbe3a介导急性心肌梗死后的心肌纤维化

这项研究旨在探讨环状RNA(circRNA)在M2巨噬细胞(M2M)衍生的小细胞外囊泡(SEVs)中在心肌纤维化发展中的作用。方法:在小鼠急性心肌梗死模型中评估了M2M衍生的细胞外囊泡(EVs)的调节作用。免疫荧光,定量实时PCR(RT-qPCR),纳米颗粒示踪分析,蛋白质印迹分析和电子显微镜用于鉴定巨噬细胞,大细胞外囊泡(LEV)和SEV。通过微阵列分析确定了M0巨噬细胞(M0Ms)和M2Ms的circRNA表达谱。进行了生物信息学分析,细胞共培养和细胞增殖测定,以研究circUbe3a在体外的表达、功能和调控机制。使用qPCR、RNA免疫沉淀(RIP)、双荧光素酶报告基因、RNA荧光原位杂交(RNA-FISH)、Western印迹分析和一系列拯救实验来验证circUbe3a,miR-138-5p和RhoC之间的相关性。结果:M2M衍生的SEV的CircUbe3a触发了心脏成纤维细胞(CFs)的功能改变。在心肌梗死后M2M渗透增加的情况下将CircUbe3a加载到SEV中。释放的SEV与质膜的融合可能导致circUbe3a释放到CF的胞质中。沉默或过表达circUbe3a在体外改变了CF的增殖、迁移和表型转化。我们证实,circUbe3a通过增强吸附miR-138-5p,然后在体外抑制RhoC翻译,在增强CF的功能变化中起着至关重要的作用。在体内,M2M衍生的SEV的添加或circUbe3a的过表达显著加剧了急性心肌梗塞后的心肌纤维化,而circUbe3a特异的shRNA则部分消除了这些作用。结论:我们的发现表明,M2M衍生的circUbe3a SEV通过直接靶向miR-138-5p / RhoC轴来促进CF的增殖,迁移和表型转化,这也可能加剧急性心肌梗塞后的心肌纤维化。

胶质瘤是最常见和致命的原发性恶性脑肿瘤。胶质瘤干细胞(GSC)可能是胶质瘤细胞增殖、侵袭、放化疗耐受和复发的重要因素。因此,发现新的GSCs相关的环状RNA(circRNA)可能找到治疗神经胶质瘤的预期目标。我们的研究中首次发现了一种新型的circRNA-CHAF1A(circCHAF1A)。 CircCHAF1A在神经胶质瘤中过表达,与低存活率有关。从功能上,发现无论在体外还是体内,circCHAF1A都能促进TP53wt GSC的增殖和肿瘤发生。从机制上讲,circCHAF1A吸附miR-211-5p上调了GSC中转录因子HOXC8的表达。然后,HOXC8可以转录上调MDM2的表达并抑制p53的抗肿瘤作用。此外,RNA结合蛋白FMR1可以通过维持其稳定性而与circCHAF1A结合并促进其表达,而HOXC8还转录FMR1的表达以形成反馈环,这可能与神经胶质瘤的恶性转化有关。 GSC中FMR1,circCHAF1A,miR-211-5p和HOXC8之间的新型反馈环可以促进神经胶质瘤和GSC的增殖和肿瘤发生。它还为神经胶质瘤的诊断和预后评估提供了有用的生物标志物,可用于分子靶向治疗。
3、 CircRNA VIM沉默与七氟醚协同作用,通过同时调节miR-124 / PD-L1轴来抑制食道癌的免疫逃逸和多种致癌活性

背景:环状RNA(circ-VIM)是急性髓细胞白血病预后不良的预测指标,但我们对其在食道癌(EC)中的功能了解甚少。在这里,我们检查了circ-VIM与七氟醚一起对EC的免疫逃逸和多种致癌活性的影响。方法:使用生物信息学工具、荧光素酶报告基因和RNA免疫沉淀法检测circ-VIM、miR-124-3p(miR-124)和PD-L1之间的调控。 CCK-8、划痕愈合和Transwell实验分别用于测量细胞增殖、迁移和侵袭。分别使用LDH分析、CFSE染色和Annexin V / PI染色检查EC细胞对CD8 + T细胞的细胞毒性、增殖和凋亡的影响。使用异种移植模型和EC细胞的尾静脉注射评估了体内肿瘤发生和肺转移。结果:在EC组织或细胞中检测到circ-VIM和PD-L1的显著上调以及miR-124的下调。 Circ-VIM吸附miR-124,并释放了其对下游目标PD-L1的抑制作用。独立于circ-VIM的七氟醚也上调了miR-124,从而降低了PD-L1的表达。通过调节miR-124 / PD-L1轴,沉默circ-VIM并应用七氟醚在体外可抑制EC的免疫逃逸和多种致癌活性,并在体内抑制异种移植物的生长和肺转移。 Ras / ERK信号通路的失活通过沉默circ-VIM和七氟醚治疗参与了恶性表型的抑制。结论:分别通过调节miR-124 / PD-L1轴来沉默circ-VIM和应用七氟醚具有协同效应,可抑制EC细胞的免疫逃逸和多种恶性表型。
