标题:Circular RNA circHIPK3 promotes homeostasis of the intestinal epithelium by reducing miR-29b function

环状RNA(circRNA)是一类形成共价闭合环的内源性非编码RNA。尽管 circRNA 影响许多生物过程,但对其在肠上皮稳态中的作用知之甚少。我们调查了维持肠上皮完整性所需的 circRNA,并确定 circHIPK3 是急性损伤后肠上皮修复的主要调节因子。方法上:从暴露于盲肠结扎穿孔(CLP)48 小时的小鼠和患有炎症性肠病(IBD)和败血症的患者中收集肠粘膜组织。我们从小鼠的小肠中分离出原代肠上皮细胞并衍生出肠道类器官。通过转染靶向circHIPK3环化位点的 siRNA 或使用过表达 circHIPK3 ,在体外损伤模型中检查肠上皮修复。 circHIPK3与miR-29b的关联通过生物素标记的RNA pulldown 测定。结果表明:全基因组图谱分析确定了大约 300 个相对于假手术组小鼠,在 CLP小鼠的肠粘膜中差异表达 circRNA,包括 circHIPK3。IBD和败血症患者的肠黏膜 circHIPK3 水平降低。增加 circHIPK3 水平可增强受伤后肠上皮修复,而circHIPK3沉默抑制上皮恢复。 circHIPK3沉默还抑制了IECs和肠道类器官的生长,circHIPK3过表达促进了小鼠肠道上皮的更新。机制研究表明,circHIPK3 直接与 miR-29b 结合并抑制 miR-29 活性,从而增加受伤后 IEC 中 Rac1、Cdc42 和细胞周期蛋白 B1 的表达。在对小鼠、IEC 和人体组织的研究中,我们的结果表明 circHIPK3 至少部分通过降低 miR-29b 的可用性来改善肠上皮的修复。

最近的研究表明,环状RNA(circRNA)在包括乳腺癌(BC)在内的多种癌症的发生和发展中发挥着重要作用。然而,大多数circRNA的潜在功能和BC进展的分子机制仍然难以捉摸。该研究中,Circular RNA 微阵列技术检测4对乳腺癌组织和癌旁组织。通过qRT-PCR和原位杂交确定circACTN4在BC细胞和组织中的表达和预后意义。实施功能获得和缺失实验以观察circACTN4对体外和体内BC细胞生长、侵袭和转移的影响。在机制上,进行了染色质免疫沉淀、荧光素酶报告基因、RNA pulldown、质谱、RNA 免疫沉淀、荧光原位杂交和免疫共沉淀分析。结果显示:CircACTN4 在乳腺癌组织和细胞中显著上调,其表达与 BC 患者的临床分期和不良预后相关。 circACTN4的异位表达显著促进了体外和体内乳腺癌细胞的生长、侵袭和转移。而敲低 circACTN4 则显示出相反的作用。 CircACTN4 主要分布在细胞核中。进一步的机制研究证明,circACTN4可以与远上游元件结合蛋白1(FUBP1)竞争性结合,阻止FUBP1与FIR结合,从而激活MYC转录,促进乳腺癌的肿瘤进展。此外,我们发现上游转录因子 2(USF2)可能促进 circACTN4 的生物发生。我们的研究结果揭示了一个关键机制,即USF2介导的circACTN4可能与FUBP1相互作用,通过增强MYC的表达促进乳腺癌的发生和发展。 CircACTN4 可能成为诊断和治疗乳腺癌的新潜在靶点。文献解读请点击:CircACTN4促进乳腺癌发生发展的机制研究
