在过去十年中,环状RNAs(CircRNAs)已成为一大类主要的非编码RNA分子,其中许多通过不同的作用机制在癌症的发展和进展中发挥关键作用。CircRNAs通常具有组织和癌症特异性表达模式,积累的数据表明这些分子具有潜在的临床相关性和实用性。尤其是,CircRNAs作为诊断、预后和预测性生物标记物具有强大的潜力,其在血浆、唾液和尿液等液体活检样本中的可检测性突显了这一点。然而,需要解决CircRNAs检测和评估中的技术问题以及生物学知识缺口,以推进这一相对年轻的研究领域,并将CircRNAs推向临床实践的前沿。在此,作者回顾了目前关于circRNAs在癌症中的生物发生、调节和功能的知识,以及它们作为生物标记物、治疗剂和药物靶点的临床潜力。
CircRNAs在肿瘤中的作用机制

背景:环状RNAs在肿瘤发生和发展中起着重要作用,但在鼻咽癌患者中尚未得到充分研究。
方法:通过RNA测序分析在NPC细胞中筛选环状RNA circCAMSAP1。通过实时定量聚合酶链反应(RT-qPCR)和原位杂交检测circCAMSAP1在鼻咽癌组织中的表达。采用创伤愈合、transwell、MTT和流式细胞术检测以及裸鼠肿瘤模型,在体外或体内研究 circCAMSAP1对鼻咽癌增殖和转移的影响。用质谱法筛选circCAMSAP1调控的下游蛋白。使用环状RNA免疫沉淀法和荧光素酶报告分析鉴定 circCAMSAP1和SERPINH1 mRNA之间的相互作用。SERPINH1和转录因子c-Myc之间的相互作用通过共免疫沉淀(Co-IP)和免疫荧光进行了验证。通过RT-qPCR和染色质免疫沉淀检测c-Myc对 circCAMSAP1产生的影响。最后,通过RT-qPCR和RNA免疫沉淀(RIP)研究了促进 circCAMSAP1产生的剪接因子。
结果:研究发现circCAMSAP1在鼻咽癌组织中高表达,促进了鼻咽癌的增殖和转移。此外,circCAMSAP1通过与SERPINH1的3′非翻译区(3′UTR)结合来提高SERPINH1 mRNA的稳定性,从而促进SERPINH1的表达。高表达的SERPINH1降低了c-Myc的泛素化降解率,导致肿瘤发生增加。同时,c-Myc与剪接因子10(SRSF10)协同作用,也能促进CAMSAP1前体mRNA转录和后剪接,形成circCAMSAP1产生的正反馈,导致鼻咽癌的增殖和转移。
结论:研究结果显示,circCAMSAP1通过结合SERPINH1的3’UTR促进NPC的增殖和转移,这表明circCAMSAP1-SERPINH1-c-Myc的正反馈可作为NPC患者的预后生物标志物或治疗靶点。

3. 实体瘤中不同环状RNA特征的表征

背景:环状RNAs(CircRNAs)在正常组织和癌组织中有差异表达,与肿瘤的发生和发展有关。然而,不同癌症类型中失调的CircRNAs的综合情况尚不清楚。
方法:在本研究中,作者对肿瘤组织及其邻近的正常样本(包括胶质母细胞瘤、食管鳞状细胞癌、肺腺癌、甲状腺癌、结直肠癌、胃癌和肝细胞癌)进行了核糖体转录组测序。CIRCexplorer2用于鉴定CircRNAs和失调的CircRNAs,基因由DESeq2包确定。通过逆转录和定量实时PCR检测hsa_circ_0072309(CircLIFR)的表达,并通过Transwell和伤口愈合实验检测其对细胞迁移的影响。通过尾静脉注射和脾脏注射分别用于肺和肝转移的小鼠模型,评估了CircLIFR在肿瘤转移中的作用。
结果:在七种实体瘤中发现了不同的circRNA表达特征,失调的circRNA表现出癌症特异性表达或在癌症中具有共同的表达特征。生物信息学分析表明,宿主基因和/或RNA结合蛋白的异常表达导致癌症中的环状RNA失调。最后,实验证实,在六种实体瘤中,CircLIFR表达下调,在体外显著抑制细胞迁移,在体内显著抑制肿瘤转移。
结论:研究结果为实体瘤中差异表达的CircRNAs提供了一个全面的图景,并强调CircRNAs广泛参与了癌症的发病机制。
